GPR56 contains seven transmembrane domains and a mucin-like domain in the N-terminal region. GPR56 is widely distributed, with highest levels found in the thyroid gland, brain, and heart. It is also expressed in a great number of tumor cells. GPR56 could be involved in cell-cell interactions. GPR56 is implicated in the regulation of brain cortical patterning. It binds specifically to transglutaminase 2 in the extracellular space. Its expression is down-regulated in melanoma cell lines, and over-expression of GPR56 can suppress tumor growth and metastasis. Mutations in GPR56 result in bilateral frontoparietal polymicrogyria.
